A highly sensitive assay for prion detection

چکیده
*Correspondence to: Ryuichiro Atarashi; Email: [email protected] We recently developed a new in vitro amplification technology, designated “real-time quaking-induced conversion (RT-QUIC),” for detection of the abnormal form of prion protein (PrP) in easily accessible specimens such as cerebrospinal fluid (CSF). After assessment of more than 200 CSF specimens from Japanese and Australian patients, we found no instance of a false positive, and more than 80% accuracy for the correct diagnosis of sporadic CreutzfeldtJakob disease (sCJD). Furthermore, the RT-QUIC can be applied to other prion diseases, including scrapie, chronic wasting disease (CWD) and bovine spongiform encephalopathy (BSE), and is able to quantify prion seeding activity when combined with an end-point dilution of samples. These results indicate that the RT-QUIC, with its high sensitivity and specificity, will be of great use as an early, rapid and specific assay for prion diseases.

متن کاملA Simple, Versatile and Sensitive Cell-Based Assay for Prions from Various Species
Detection and quantification of prion infectivity is a crucial step for various fundamental and applied aspects of prion research. Identification of cell lines highly sensitive to prion infection led to the development of cell-based titration procedures aiming at replacing animal bioassays, usually performed in mice or hamsters. However, most of these cell lines are only permissive to mouse-ada...
متن کاملGold nanoparticles as a probe for prion determination via resonance light scattering method.
Dihydrolipoic acid (DHLA) modified gold nanoparticles (AuNPs) were used as a highly selective probe for the detection of prion proteins. We discovered that AuNPs undergo aggregation selectively in the presence of recombinant prion protein (rPrP), and such selective aggregation enhanced the resonance light scattering (RLS) intensity from AuNPs tremendously. Based on this phenomenon, we establish...

متن کاملRapid End-Point Quantitation of Prion Seeding Activity with Sensitivity Comparable to Bioassays
A major problem for the effective diagnosis and management of prion diseases is the lack of rapid high-throughput assays to measure low levels of prions. Such measurements have typically required prolonged bioassays in animals. Highly sensitive, but generally non-quantitative, prion detection methods have been developed based on prions' ability to seed the conversion of normally soluble proteas...

Real-time quaking-induced conversion (RT-QuIC), a highly specific and sensitive assay able to detect low levels of the disease-inducing isoform of the prion protein (PrP(d)) in brain tissue biopsies and cerebral spinal fluid, has great potential to become a method for diagnosing prion disease ante mortem. In order to standardize the assay method for routine analysis, an understanding of how phy...
